I use a three-pronged approach. AdAware (as listed above), as well as Spybot (Search %26 Destroy), which catches some things AdAware doesn't and has a built in blocker to keep it out for good, and Spywareblaster, which is purely a blocker.

Lavasoft's Adaware is great, so is AVG Antivirus.
Use Mozilla Firefox with the pop-up blocker. It is even better than IE, though some sites don't allow.
For anti-spywares go to download .com and check out all the free downloads. It is more effective when you have multiple anti-spywares in your PC.
Ad-aware works best for me
